Effects of Imipramine to the Fetus and the Pregnant Rats

Abstract
Histological changes of pituitary glands caused by the administration of imipramine hydrochloride during the gestation period of the rat were studied immunohistochemically using the enzyme-labeled antibody.
Eighth to twelfth days after injection of 40mg. (per kg of body weight) of imipramine rats were sacrificed by decapitation and the pituitary glands were carried out to make ordinary histological sections. Prolactin granules were reacted with rabbit anti-ovine prolactin and peroxidase labeled sheep anti-rabbit gamma globulin successively and the peroxidase were localized histochemically.
Prolactin cells from the rats which was given imipramine showed marked loss of granules as well as the lowering of intensity to the histochemical reaction.
The drug seems to effect on the hypothalamus and so to the pituitary glands somewhat longer period, while imipramine did not induce the abortion or the other failure of the gestation when the drug was given during the sixth to twelfth days of gestation.
